Procollagen-proline dioxygenase (also known as protocollagen hydroxylase, prolyl hydroxylase, prolyl 4-hydroxylase and protocollagen prolyl hydroxylase) is an enzyme involved in collagen synthesis. It catalyzes the formation of hydroxyproline via the following, Vitamin C-dependent reaction:

procollagen L-proline + 2-oxoglutarate + O2 → (2S, 4R)-4-hydroxyprolinesuccinate + CO2

Some prolyl hydroxylases also act on hypoxia-inducible factors (HIFs) which are transcription factors that can modulate cellular metabolism under conditions of decreased oxygen.
